What are high purity solvents? And why do they matter so much in the chromatography process? 

Chromatography is the separation of analytes from a mixture, using:

1. A stationary phase, in which the analyte separates and is stored 
2. A mobile phase, to separate the analytes from the stationary phase

Solvents play an essential role as the components of the mobile phase, helping transport the sample to the detector, after passing through the separation column. Analytes with less affinity for the stationary phase will elute earlier, while those with greater affinity will elute later to achieve separation. The solvents used for the mobile phase (and the ration used) can be adjusted to change the relative analyte affinity – which changes the retention time and selectivity of the separation. To predict the elution order of analytes, you need to know the type of chromatographic column used, and the polarity of the mobile phase solvents. 

Not all solvents are created equal. GC and HPLC solvents are available commercially under different purity grades, to meet different analysis requirements. HPLC analysis requires the highest purity HPLC grade solvents. The purity of the solvent is extremely important when it comes to quality control, as it can affect the achievable sensitivity of the chemical analysis. 

How to choose the right HPLC solvent for your application?

• UV Cut-off: No solvent is 100% transparent over the entire UV-visible spectrum, but HPLC solvents show transparency up to a certain wavelength range, below which they absorb. 
• Viscosity: Aim for low viscosity in HPLC grade solvents. The higher the viscosity, the higher the back pressure, due to resistance by the stationary phase in the packed column. 
• Stability: Solvents should be stable in sealed solvent bottles, but be sure to follow storage instructions and check their shelf life prior to use, to ensure staff and laboratory safety.
